چکیده

VOLATILE anesthetics are well known to have cardioprotective effects similar to those of ischemic preconditioning, but the mechanisms remain unclear. Recent evidence indicates that the protective effects of KATP channel openers may be due to action on mitochondrial KATP channels. 12–16 We have previously reported that 15 min of volatile anesthetic exposure induces mitochondrial flavoprotein oxidation via mitochondrial KATP channel activation, which may contribute to anesthetic-induced preconditioning. Volatile anesthetic exposure produces a memory period after exposure during which damage from ischemia is diminished, even though the anesthetic is no longer being administered. We designed the present study to examine time factors that may influence mitochondrial oxidation in anesthetic-induced preconditioning using mitochondrial flavoprotein fluorescence (MFF) as an endogenous reporter of mitochondrial redox state.

Isoflurane preconditioning uncouples mitochondria and protects against hypoxia-reoxygenation.

Ischemic cardiac injury can be substantially alleviated by exposing the heart to pharmacological agents such as volatile anesthetics before occurrence of ischemia-reperfusion. A hallmark of this preconditioning phenomenon is its memory, when cardioprotective effects persist even after removal of preconditioning stimulus. Since numerous studies pinpoint mitochondria as crucial players in protect...

Isoflurane alters energy substrate metabolism to preserve mechanical function in isolated rat hearts following prolonged no-flow hypothermic storage.

BACKGROUND Isoflurane enhances mechanical function in hearts subject to normothermic global or regional ischemia. The authors examined the effectiveness of isoflurane in preserving mechanical function in hearts subjected to cardioplegic arrest and prolonged hypothermic no-flow storage. The role of isoflurane in altering myocardial glucose metabolism during storage and reperfusion during these c...

Dual effects of isoflurane on proliferation, differentiation, and survival in human neuroprogenitor cells.

BACKGROUND Previous studies have demonstrated that isoflurane can provide both neuroprotection and neurotoxicity in various tissue culture models and in rodent developing brains. The cellular and molecular mechanisms mediating these dual effects are not clear, but the exposure level and duration of isoflurane appear to be determinant factors. METHODS Using the ReNcell CX (Millipore, Billerica...

Effects of tail fat on recovery times of anesthesia with isoflurane in fat-tailed Iranian Lori-Bakhtiyari lambs

In the present study, the effect of tail fat on recovery times in intact sheep and sheep with a ligated median sacral artery following similar anesthetic exposure with isoflurane was investigated. This study was performed using seven healthy fat-tailed Iranian Lori-Bakhtiyari ewe lambs. The lambs were anesthetized twice at two week intervals (the experiment was performed in two stages). After m...

Isoflurane reduces hypoxia/reoxygenation-induced apoptosis and mitochondrial permeability transition in rat primary cultured cardiocytes

BACKGROUND The volatile anesthetic isoflurane protects the heart from hypoxia/reperfusion (H/R) injury. However, it is still incompletely understood whether isoflurane exerts its protective role through preventing mitochondrial permeability transition pore (MPTP) opening. METHODS Primary cultured cardiocytes were exposed to H/R in the absence or presence of isoflurane. Cell cytotoxicity and a...
